If someone who you think you might want to take things further with tells you that they like your anklet, how do you react?

I would probably just say thank you, generally open up my body language to show receptivity (smile turn, to face him) and ask him a question or make an observation. I typically do not jump right into any kind of explicit comment or discussion of what the anklet means. It is more a matter of having a conversation to see if I am interested. If he asks me if it means what he thinks it means I will play along, but probably be a bit coy in a playful positive way. Something like "what do you think it means?" or "yes it probably does".

While there is an obvious sexual overtone to the hotwife anklet, I view my interactions with men as being much like it was when I was single. While my attitude and approach to sexual engagement has evolved from when I was single, I am still discriminating and want to take the time to see if a guy genuinely appeals to me.

In some cultures/lifestyles it may mean something. For the vast majority of people it's just jewelry.

Exactly correct, I'm guessing you're Indian like me so an anklet for Indian women is to ward off the evil eye.
